Experimental outline. ATAC-seq maps accessible regions in the genome by taking advantage of the preference of the Tn5 transposase for inserting into physically accessible DNA. We mapped 5-hmU using two orthogonal approaches — MeDIP-seq, using an α 5-hmU antibody, and chemical conversion, using oxidation of 5-hmU to 5-fU, eventually resulting in T-to-C base conversion in final libraries.
