The AIM peptide of Rab3GAPL (Rab3GAPL-AIMp) directly binds to ATG8CL in vitro. The binding affinity of Rab3GAPL-AIMp to ATG8CL was determined using isothermal titration calorimetry (ITC). The upper panel shows heat differences upon injection of Rab3GAPL-AIMp into ATG8CL and the bottom panel shows integrated heats of injection and the best-fit line to a single site binding model. KD= 962 nM,N= 1.03. In the sequence illustration, Rab3GAPL-AIMp is represented in black color, with key AIM residues highlighted in pink for WT Rab3GAPL-AIMp, and in yellow for the mutated version.
