In SMRT-Tag, hairpin adapter-loaded Tn5 transposase was used to fragment DNA into kb-scale fragments. The 9-nt gaps introduced by transposition were closed via optimized gap repair, while exonuclease digestion enriched for the covalently closed templates required for PacBio sequencing.
