K562 cells piggyBac-engineered to constitutively express the indicated panel of fusion protein constructs were transduced with one of four 3-plex crRNA constructs targeting the MYC promoter or cotargeting the three enhancers using one crRNA per enhancer. Cell fitness as a proxy of MYC expression is measured as log 2 fold-change in percentage of cells expressing GFP marker on the crRNA construct, relative to day 6 after crRNA transduction. The averages of two biological replicates ( n = 3,4328,872 cells per replicate) are shown as individual data points and the range denoted by vertical lines.
