To resolve this dropout problem, a constitutively and highly expressed Pol III-derived circularized barcoded RNA 34 (Tornado barcodes, oBC, blue), a priori matched with the mBC (red) and CRE, is appended co-directionally upstream in a dual RNA cassette. The oBC enables robust detection of reporters in single cells, independent of reporter activity, enabling unbiased measurement of mBCs from the CRE-driven reporter mRNA.
