Translocation of SPG20 and recruitment of ITCH to damaged lysosomes depends on bulky hydrophobic residues in the SC domain of SPG20. SPG20 KO cells expressing GFP-tagged versions of SPG20 wt, SPG20 8xV, or SPG20 8xV fused to the ALPS motif of ARFGAP1 (8xV-ALPSARFGAP1) were mock or LLOMe-treated and costained for ITCH and LAMP1 (not shown here). See Figure S8H for control treatments.
