Lysophagy assay. HeLa cells stably expressing pH-sensitive mKeima fused to TMEM192. Due to its position on the cytosolic side of the lysosomal membrane, mKeima is acidified and shifts its excitation maximum from green to red during lysophagy but not during lysosome repair. Cells transfected with indicated siRNAs and treated with LLOMe followed by washout. Scale bars: 10 um.
