C6 interacts directly with HDAC5 via the N-terminal domain. TAP-tagged N1 or C6 was co-expressed with HA-tagged HDAC1, HDAC5, or H5N using a wheat germ cell-free transcription and translation system. Left and center left: HA-tagged proteins were immunoprecipitated, and the purified proteins were analyzed by immunoblotting using antibodies as indicated. Right and center right: the same co-expression samples were prepared without N1, TAP-C6 was precipitated, and purified proteins were analyzed as described above. In all immunoblots, the positions of molecular mass markers are shown in kiloDaltons on the right.
