Schematic of derivation of embryonic stem cells (ESCs) from chimeric embryos. PGCs are isolated from E8.5E9.5 embryos by FACS and injected into host embryos at 8-cell stage. After 48 h of in vitro culture, trophectoderm cells are removed by immunosurgery, and primary colonies are then cultured in 2i/LIF, then expanded by passaging.
