Targets are labeled with a primary (1ry) DNA barcode (red) and a secondary (2ry) label composed of the full barcode complement sequence, a toehold (orange) and the speed-optimized DNA-PAINT docking sequences (gray). For readout, a complementary dye-labeled imager strand is added.
