Workflow from Scientific Research

Molecular approach. Two sequencing libraries are prepared from the same set of barcoded nuclear cDNAs. The first is a conventional snRNA-seq library. The second library samples the CAG-repeat sequence in HTT gene transcripts and is analyzed by long-read sequencing. The presence of shared cell barcodes in the two libraries allows each CAG-repeat sequence to be matched to the RNA-expression profile of the nucleus from which it was sampled.
