Generation of G188R rhodopsin knockin mice by CRISPR/Cas9 gene targeting. Sequence in the region of the mutation are shown. sgRNA was selected so that Cas9 would cut in exon 3 in the vicinity of glycine (GGG) at position 188. Homology-directed repair in the presence of a targeting oligo with the desired mutation introduces an arginine (AGA) mutation at position 188. Chromatograms in the region of the mutation are shown from sequencing of genomic DNA from B6 and Rho G188R mice.
