Model of lipid droplet (LD) biogenesis. Neutral lipids such as triglycerides accumulate within ER membranes, leading to leaflet displacement (lens). The LD grows outward from the cytosolic face of the ER, eventually forming a distinct droplet encapsulated by a phospholipid monolayer. LDs can form special contacts sites (indicated with an arrow) that allow them to remain connected to the ER. They also form regular contact sites with other organelles.
