Genetic scheme for type II lineage filtering using the CLIn technique. A type-II-specific promoter fragment (stg-14) combined with KD recombinase removes a STOP sequence by a flip event and leaves Cre recombinase active in type II NSCs. Cre generates type II NSC clones positive for LexA::p65. GAL80 is active in the rest of the cells, and GAL4 expresses GFP as a reporter in class-specific manner. We did not stain for mCherry here.
